报告摘要:
Microalgae produce large amounts of triacylglycerols (TAGs) under nitrogen deprivation, which can be used as biodiesel feedstocks. Genes required for TAG synthesis are promising targets to be engineered towards enhanced biofuel production. To identify such genes, together with colleagues at Michigan State University, I performed a high-throughput screen for mutants with abnormal TAG content in the model green alga-Chlamydomonas reinhardtii. One mutant, pgd1, exhibited 50% lower TAG content compared to the wild-type. Mechanistic studies suggest that PGD1 functions as a galactoglycerolipid lipase that facilitates export of fatty acids from the chloroplast. Over-expression of PGD1 to enhance TAG production is in progress.
At Carnegie Institution for Science, we (the mutant library team in the Jonikas Lab) aim to generate the first genome-wide indexed insertional mutant library for Chlamydomonas. We developed genomic tools for mapping mutation sites as well as robotic approaches for mutant generation, propagation and cryopreservation. We have generated a pilot scale mutant library that covers 8% Chlamydomonas genes with high-confidence. A 10X larger scale library has been generated and is being mapped. This resource will transform genetic studies in microalgae.
Besides genetic interrogations of TAG synthesis pathways, we are also interested in understanding the physiological significance of TAG production. Preliminary evidence suggests a role of TAG to serve as an electron sink under nitrogen deprivation to prevent oxidative burst since pgd1 cells die during nitrogen deprivation due to redox imbalance. We developed a tool for pooled screen to obtain all mutants that cannot survive nitrogen deprivation. This will shed light on how photosynthetic organism survive nutrient deprivation as well as providing more TAG deficient mutants.
